CLINICAL STUDY Estrogen receptor a gene polymorphisms (Pvu II and Xba I) influence association between leptin receptor gene polymorphism (Gln223Arg) and bone mineral density in young men

چکیده
Objective: The peak bone mass is recognized as an important determinant in the development of osteoporosis. We investigated associations between bone mineral density (BMD) and polymorphisms of the leptin receptor (LEPR) and estrogen receptor a (ERa) genes in young men. Design: BMD, anthropometric characteristics, and serum leptin concentrations were measured in young men and compared with regard to the LEPR and ERa genotype. Methods: From 219 healthy volunteers aged 20 –34 years, we genotyped the Lys109Arg, Gln223Arg, Ser492Thr, Ala976Asp, and Pro1019Pro variants of LEPR and the Pvu II and Xba I variants of ERa using the polymerase chain reaction-restriction fragment length polymorphism method. We determined serum leptin concentrations by radioimmunoassay (RIA) and BMD by dual energy X-ray absorptiometry. Results: The subjects carrying the Gln223 allele of LEPR had higher BMD at the lumbar spine compared with the subjects without this allele. There were no significant differences in BMD among Pvu II and Xba I genotypes of ERa. However, an association between LEPR and BMD was noted in the subjects carrying the PP homozygotes of Pvu II or the X alleles of Xba I, but this was not significant in those without these genotypes. Conclusions: This study indicates that the Gln223Arg polymorphism of LEPR is important for determination of the peak bone mass in men and that it is influenced by ERa gene polymorphisms. P-238: Lack of Association of Estrogen Receptor Polymorphisms with Male Infertility
Background: Estrogen is recognized as one of the significant regulator of spermatogenesis. Estrogens are synthesized in the male reproductive system (sertoli and leydig cells). Estrogen function is mediated by estrogen receptors (ER-α or ER-β). Some studies have suggested an association between single nucleotide polymorphisms (SNPs) rs2234693 (ESR1 pvuII C>T), rs1801132 (ESR1 325 C->G) of ERα g...
متن کاملPolymorphisms in the gene encoding estrogen receptor alpha are associated with osteoarthritis in Han Chinese women.
Polymorphisms in the Xba I and Pvu II restriction enzyme recognition sites in the estrogen receptor-alpha gene (ESR1) have been associated with multiple diseases, including osteoarthritis. To determine whether such polymorphisms are associated with osteoarthritis in a Han Chinese population, 98 women with osteoarthritis and 196 healthy women were genotyped by PCR-RFLP of ESR1 with Xba I and Pvu...
متن کاملAssociation of ESRα Gene Pvu II T>C, XbaI A>G and BtgI G>A Polymorphisms with Knee Osteoarthritis Susceptibility: A Systematic Review and Meta-Analysis Based on 22 Case-Control Studies
Background: Many studies have reported the association of estrogen receptor α gene (ESRα) ESRα PvuII T>C, XbaI A>G and BtgI G>A polymorphisms with Knee osteoarthritis (KOA) risk, but the results remained controversial. In order to drive a more precise estimation, the present systematic review and meta-analysis was performed to investigate the association between ESRα polymorphisms and KOA susce...
متن کاملLipid Profile and Leptin Levels in Patients with Metabolic Syndrome
Abstract Background and Objective: Metabolic syndrome called a cluster of several metabolic disorders is associated with increased risk of cardiovascular diseases. Genetic differences in leptin receptor gene are related with the concentration and activity of leptin in that these discrepancies can influence lipid levels. We aimed to determine the association between the leptin receptor gene p...
متن کاملبررسی ارتباط پلی مورفیسم Fok1 گیرنده ویتامین D3 با تراکم استخوان در زنان یائسه مبتلا به استئوپروز
Background and purpose: Osteoporosis is a multiple disease that is characterized by decrease in bone mass and bone strength. This decrease is more common in postmenopausal women due to decreased estrogen levels. Vitamin D3 plays a pivotal role in skeletal metabolism by binding to nuclear steroid receptors (receptors for vitamin D (VDR)). This study aimed at investigating the relationship betwee...
